Hi,

You need to set the "Scan resolution" on the Input Tab to 4000 PPI, and the Bits per pixel setting to 48 bit RGB. On the Crop tab, you could set the Crop size to Maximum (or place the Crop boundaries yourself in the preview). Then on the Output tab, you set the Printed size to Scan size, Magnification to 100%, make sure you have selected Tiff file as output, Tiff size reduction is 1, and Tiff file type is 48 bit RGB.

Cheers,
Bart
