A couple of profiling questions

When profiling glassy papers for the 3800, if I'm setting output levels at, say, 245 for the purposes of limiting gloss differential, should I redo my paper profiles with this setting on the targets?

Also;

for the purposes of profile testing only, can I use "convert to profile" with printer profiles and print the resulting file with no printer or photoshop color management, thus enabling me to test different profiles on a larger sheet of paper with only one pass through the printer?

For your first question I'd say yes. If you change anything related to the ink settings you should generate a new profile.
