I never Set up a Camera Profile. Also, does it make a significant difference using Camera Profile? I am using Pentax 645 Z
Is there any tutorial that will explain this to me? Thank you

A lot of tutorials on Youtube.

Yes it makes a difference and is recommended.

If you're shooting with a 645z, you should download the DNG profile Torger created. By far the best I have seen for this camera.
Do a search within the Color Management folder on "dcamProf" and "654z"
Good luck!
