Spydercheckr 48 low light

Hi there,

I recently purchased the great bundle from Datacolor, which includes the Spydercheckr 48 grid.
I am new to this kind of tool and I have a few questions:

1- Wouldn't it be enough to make one calibration per lens/body configuration and then apply it to every picture taken with that combination, as long as the picture's white balance is neutralized (with the help of a grey card on location)?

2- If not, then in the case of a low light condition shoot (night photography, canyons, caves...) how can you make an appropriate calibration without setting ridiculously long shutter speeds or high ISO (the white point square on the checker needs to be 96% RGB for accurate colour correction, according to the manufacturer).

I hope these questions make sense, and that someone will come up with an answer, as I plan on using these tools for my next trip coming up quite quickly!
