I have just put together a system consisting of:
i6800k
64GB 3000Mhz RAM
GTX 1070 GPU
ASUS X99 MB
The RAM is currently running at the MB default speed of 2133.
On this MB, faster RAM speeds can only be achieved by overclocking.
Should I tweak the system? Would I see an appreciable difference?
If so, what should I do in BIOS?
Or should I leave well enough alone?

Make sure your entire workflow is with NVMe and SSD drives. Otherwise, all this hardware will be waiting for data to load.

Yes.
OS and CS6 are on one fast SSD drive. Files will be imported onto another SSD for editing, before being stored on SATA drives.
Any BIOS tweaks?

Why not?
Just need to keep the CPU cool and not abuse the voltages.
I would not use CPU Core Voltage more than 1.3 volts.
4ghz/4.1Ghz is fine and will not generate too much heat if you got
a decent water or TOP air cooler.

The Asus over clocking utility will get you there quickly and safely. Good cooling is needed to over clock or CPU internal safeties will defeat any speed/load you try to reach.

Cooler is a NZXT Kraken X61.
Since changing to XMP, it's a Porsche with a keyboard. RAM is now 3000Mhz.
Am very happy with it.
Real test for me will be video rendering.
